Comparison of Two Methods for Purifying the Rabbit Antibody of Acid Red 73 IgG

Abstract: To compare the effects of saturated ammonium sulfate method and caprylic acid-ammonium sulfate method for purifying the rabbit anti-AR73 IgG. The IgG were tested in protein content,relative molecular weight, the antibody titer by BCA method,sodium dodecyl sulfate polyacrylamide gel electrophoresis (SDS-PAGE) and indirect enzyme linked immunosorbent assay (ELISA).The results indicated that the protein content of IgG by saturated ammonium sulfate method was 36.44 mg/mL and the antibody titer was 1.6×105, the protein content of IgG by caprylic acid-ammonium sulfate method was 15.79 mg/mL, and the antibody titer was 3.2×105. The purity of purified antibody by caprylic acid-ammonium sulfate method was higher than that by saturated ammonium sulfate method.
