Construction and Optimization Condition of Proteomic Map of Skin Collected from Inner Mongolia Cashmere Goat

Abstract: This study was aim to establish an efficient and high resolution proteomic map of skin collected from Inner Mongolia Cashmere goat. Total protein was extracted by using liquid nitrogen grinding method. The effects of different precipitate methods, different loading quantities and different gel concentration of SDS-PAGE on 2-DE (two-dimensional gel electrophoresis) maps were compared and optimized though the 2-DE. The results showed that the best electrophoresis pattern could be obtained by using 2-D Clean-up kit in protein extracting and loading 20 μg protein samples on each strip as well as using SDS-PAGE with 10% gel concentration and that would be helpful for the further proteomics analysis.
