Pseudolaric Acid B Induced Apoptosis and Senescence Through ROS in MCF-7 Cells

Abstract: To study the relationship of ROS and pseudolaric acid B (PAB)-induced MCF-7 cell apoptosis and senescence. Contrast phase microscope was used to observe cellular morphologic alternation; LDH assay was performed to assess apoptosis ratio; galactosidase staining showed cell senescence; and flow cytometric analysis of DCFH-DA staining determined the level of ROS. We found that PAB induced MCF-7 cell apoptosis in time-dependent manner. 4 μmol/L PAB treated MCF-7 for 3 d,then replace PAB with fresh DMEM for 5 d,cell became senescence. PAB increased ROS level in time dependent manner. Therefore,PAB increased ROS level to induce apoptosis and senescence.
