RAPD Analysis of the Genetic Specificity of Huainan Pig

Abstract: Genetic relationship among Duroc, Landrace and Large white were analyzed with random amplified polymorphic DNA (RAPD) technique. 150 random primers were used to amplify genomic DNA of pig with PCR method. On the basis of preliminary experiments, 72 polymorphic primers were selected to analyze the genetic variation among 4 pig lines, 911 amplified fragments were detected by agarose gel electrophoresis. Among them,462 fragments were polymorphic, accounting for 50.71% of the whole. The results showed that the RAPD polymorphisms among pig strains were abundant. The results of the statistical analysis of RAPD bands showed that the genetic similarity were 0.0068 and 0.0069 in Huainan pig and Duroc, Landrace,which indicated that Huainan pig had a closer relationship with Duroc, Landrace, respectively, and the genetic similarity between Huainan pig and Large white was 0.0072, which showed the Huainan pig were cleary apart from Large white. The amplified band patterns showed that there were both genetic similarities and genetic variations among Duroc, Landrace, Large white and Huainan pig. Thus, the RAPD technique could be used to monitor genetic relationship among different strains and different individuals of the same strain of pig.
