Detection of Forsythiaside in Plasma by HPLC

Abstract: To establish a method for the determination of forsythiaside in plasma, after dealing with the plasma protein sample,acetonitrtle-water-phosphoric acid (18∶82.5∶0.41) was taken as the mobile phase at a flow rate of 1.0 mL/min. The detective wavelength was 332 nm. The temperature was 25 ℃ and the sample volume was 10 μL. The Angilent ZORBAX SB-C18 column (5 μm × 4.6 mm × 250 mm) was taken as the column for the separation and then progresssd the chromatographic analysis. In this method the plasma concentration of forsythiaside in the glycoside 0.005 to 200 μg/mL was good linearity, precision and repeatability test RSD was less than five percent.The results showed that the method is simple, sensitive and highly reproductive, and it is suitable for the research of the pharmacokinetics and relative bioavailability of forsythiaside.
