Abstract: At the present, electroporation of skeletal muscle has been used extensively to transfer a number of functional genes in multiple large animal models. Electroporation greatly increases transfection efficiency of nonviral-vector gene in skeletal muscle. The possible mechanism was discussed on electro-gene-transfer across the cell membrane and the critical factors were analyzed which had a full impact on skeletal electro-gene-transfer efficacy include species, cell size, field strength, pulse parameters, electrodes and electroporater. 
