Production of Porcine Transgenic Embryos Expressing GFP by Intracytoplasmic Sperm Injection(ICSI)

Abstract: Intracytoplasmic sperm injection(ICSI), which is a new technique of embryo engineering of domestic animals, can solve the problem of porcine polyspermy. Porcine ICSI was conducted using repeatedly frozen/thawed intact sperm or sperm head which were incubated with GFP gene for 30 minutes.The expression rate of GFP gene in ICSI oocytes activated by CaCl2 was higher than that of ICSI oocytes activated by electrical pulse. The result showed that SMGT-ICSI procedure for the production of transgenic porcine embryos was feasible.
