Abstract: As experiments the material take the slaughter house cow ovary, studies in vitro, in vivo cultivates the method of preserving health to the external fertilization embryonic development influence. (1) IVF embryo in vitro culture system whether adds the oviduct epithelial cell monolayer (granular cell monolayer)(2×106/ml) in nutrient fluid culture and temporary acceptor sheep in vivo raise contrast experiment. Result demonstration that, In the culture 8 d BL embryo growth rate not remarkable difference, in vitro culture system BL embryo forms mainly concentrates in 8 d(fertilization date for 0 d). In vivo culture system BL embryo’s formation most in 7 d. Adds the somatic cell in vitro culture system and in vivo cultivates the method of preserving health BL embryo production rate not remarkable difference, but remarkable is actually better than not add the somatic cell simple in vitro to cultivate the method. (2)two methods to produce the embryo the procedure to freeze, the thawing sensitivity test. Result demonstration that, After in vivo cultivates the method of preserving in vivo and the ordinary in vitro law production embryo defrosts. the survival percentage (90% VS 60%) and the embryo hatching rate (85.7% VS 44.4%)have the remarkable difference (P<0.05).
