水貂多巴胺受体D1基因多态性及与自咬行为的关系

The Polymorphism of DRD1 Gene and Its Relationship with Selfbiting Behaviors in American Mink

摘要: 以多巴胺受体D1基因(DRD1)作为候选基因, 分析该基因对水貂自咬行为的影响。本试验采用聚合酶链式反应法,首次从美洲黑貂脚部肌肉组织扩增出多巴胺受体D1基因的部分序列片段,并测序,现已将该序列提交到GenBank上,得到序列号为EU249297。通过序列比较,在179位点处发生了(T→C)转换,但并没有导致氨基酸的变化。采用单链构象多态性(PCR-SSCP)方法进行了多态性检测,结果在不同个体中检测到3种基因型:AA、AB和BB型。对该基因的不同基因型与自咬行为的关系进行分析。结果表明,DRD1多态性对水貂自咬行为的影响差异显著(P<0.05)。在该群体中A等位基因的频率均高于B等位基因,AA型的基因型频率均高于BB型的基因型频率;自咬行为组AA基因型的频率远低于健康组,BB和AB基因型个体分布频率比较差异显著。 

Abstract: We report the effects of candidate gene Dopamine Receptor D1 (DRD1) gene on selfbiting in the current study. In this study, partial sequence of Dopamine Receptor D1 genes were firstly extracted from feet in American mink, and were amplified using Polymerase Chain Reaction (PCR) method, then sequenced. Now the sequence has been sent to Gene Bank with registration, and the Accession Number is EU249297. Comparison the fragment sequence of different individuals, we found T→C mutation, but coded amino acid had no change. Then followed by single strand conformation polymorphism (PCR-SSCP) analysis, there showed three kinds of genotypes: AA, AB, BB. The correlation between the genotypes with the in selfbiting behavior was analyzed, the results showed that the frequency of allele A is higher than allele B, the AA genotypes in control group were higher than that of selfbiting group, and is different significantly (P<0.05).
