鸭新城疫病毒山东株的分离鉴定

摘要: 笔者从病鸭体内分离到一株病毒,通过试验确定为新城疫病毒。参照世界动物卫生组织(OIE)规定的新城疫毒力判定的标准和方法,测定该分离株的1日龄雏鸡脑内致病指数(ICPI)和6周龄鸡静脉致病指数(IVPI)分别为1.79和2.45。采用已经发表的鸡新城疫病毒序列设计引物,应用RT-PCR技术扩增鸭源新城疫病毒的F基因,并将其克隆至载体上,然后进行了核苷酸序列、氨基酸序列测定,结果表明该分离株为新城疫强毒株。

Abstract: The virus isolated from duck was identified as Newcastle disease virus by the HA/HI. The intracerebral pathogenicity index in 1-day-old chickens, the intravenous pathogenicity 6-week-old chickens were 1.79 and 2.45 according to the standard and methods of Newcastle disease pathogenicity judgment refer to OIE. We amplified its fusion protein gene by RT-PCR using the primer which got from the reported sequence of NDV in GenBank, cloned into the carrier and sequenced. The results showed that the isolated virus was the strong virulence NDV.
