抗菌肽研发现状及其改造策略

Research Present Status and Reform Strategy of Antimicrobial Peptides

摘要: 抗菌肽已成为替代抗生素来预防和控制疾病的潜在药物,但因其产量低,生产成本高而缺乏大规模生产的条件,因此,通过基因工程技术对抗菌肽进行改造成为必然.文章分析了抗菌肽的研发现状、抗菌肽的基因改造策略,介绍了在天然抗菌肽基础上开展的氨基酸残基设计、肽片段拼接设计、肽链的环合设计、计算机辅助设计、靶向设计和模拟生物模型设计等衍生肽的分子设计策略及通过融合表达、串联表达、多拷贝表达等基因重组技术制备抗菌肽的优势及抗菌肽表达调控等研究进展,为抗菌肽的低成本和规模化生产奠定理论基础.

Abstract: Antimicrobial peptides have become potential drugs to replace antibiotic for preventing and controlling diseases,but it's still lack of suitable mass production conditions because of low yield and high production cost,so it becomes inevitable to use gene engineering technology of antimicrobial peptide reconstruction.This paper analyzed the research status of antimicrobial peptides,concluded the gene transformation strategies,such as amino acid residues design,peptide fragments junction and construction of peptide cyclization,computer aided design,the target design and simulation of biological model,and the advantage of gene recombination technology by fusion expression,tandem expression and multi copy expression.This research progress would lay theoretical foundation for low cost and large scale production of antimicrobial peptides.
