1株肠炎沙门氏菌噬菌体的污水分离及生物学特性研究

Isolation and Biological Characteristics of a Phage against Salmonella enteritidis from Sawage Water

摘要: 以1株肠炎沙门氏菌作为宿主菌,应用双层平板噬菌斑试验从污水中分离噬菌体并对其基本生物学特性进行研究。结果显示成功地从污水中分离出1株肠炎沙门氏菌噬菌体,噬菌斑呈圆形透明状,直径2 mm 左右(培养12 h),边界清楚;电镜观察结果显示该噬菌体有1个正多面体立体对称的头部,头部直径约54 nm,无囊膜,有1长尾,无收缩尾鞘,尾长约110 nm;当感染复数(MOI)为2时,子代噬菌体滴度较高;按一步生长试验结果绘制出一步生长曲线,结果显示感染宿主菌的潜伏期约为10 min,裂解期约100 min,平均裂解量约为61。该试验结果为动物沙门氏菌病的进一步噬菌体生物防制研究提供了材料。

Abstract: Using double-layer plate method, a phage against Salmonella enteritidis was isolated from sewage water, and its biological characteristics were studied. The plaques of isolated phage were transparent with clear boundary, and were about 2 mm in diameter at 12 h. The morphological properties of phage were examined by electron microscope, and showed that the phage had an isometric polyhedral no-enveloped head with diameter of about 54 nm, and a long noncontractile tail with size of about 110 nm. The test of optimal multiplicity of infection showed when MOI equaled 2, the number of phage offspring was higher. One-step growth kinetics was determined according to the results of one-step growth experiment, which showed that the latent period was about 10 min, the rise period was about 100 min, and the average bust size was about 61. The phage against S.enteritidis was successfully isolated from the sewage water in this study, which would provide materials for the further biological control researches of S.enteritidis using corresponding phage.
