摘要: 试验旨在建立内蒙古绒山羊皮肤蛋白质双向电泳图谱。采用液氮研磨法提取蛋白质,通过双向电泳技术对蛋白质进行分离,分别对不同的蛋白质沉淀方法、上样量和SDS-PAGE凝胶浓度进行比较。结果显示,使用2-D Clean-up试剂盒处理、上样量为20 μg、采用10%的SDS-PAGE凝胶浓度时可获得重复性好和分辨率高的双向电泳图谱,便于后续的内蒙古绒山羊皮肤差异蛋白质组学分析。

Abstract: This study was aim to establish an efficient and high resolution proteomic map of skin collected from Inner Mongolia Cashmere goat. Total protein was extracted by using liquid nitrogen grinding method. The effects of different precipitate methods, different loading quantities and different gel concentration of SDS-PAGE on 2-DE (two-dimensional gel electrophoresis) maps were compared and optimized though the 2-DE. The results showed that the best electrophoresis pattern could be obtained by using 2-D Clean-up kit in protein extracting and loading 20 μg protein samples on each strip as well as using SDS-PAGE with 10% gel concentration and that would be helpful for the further proteomics analysis.
