硬脂酰辅酶A去饱和酶-1基因表达及其与鸭肉质和血清生化指标的相关性分析

Correlation Analysis between SCD-1 Gene Expression and Meat Quality and Serum Biochemical Indexes of Ducks

摘要: 试验分别在基础日粮中添加含4%亚油酸和4%二十碳五烯酸(EPA)对樱桃谷鸭进行饲养,检测4、6、8和10周龄樱桃谷鸭硬脂酰辅酶A去饱和酶-1(SCD-1)基因mRNA表达丰度,分析其表达与10周龄鸭肉质和血清生化指标的相关性。实时荧光定量PCR结果显示,亚油酸和二十碳五烯酸均能显著降低各周龄SCD-1基因在鸭肝脏、脂肪和胸肌中的mRNA表达水平(P<0.05),说明亚油酸和二十碳五烯酸均具有抑制SCD-1基因转录的功能,亚油酸组鸭SCD-1基因mRNA表达水平与二十碳五烯酸组相比未达到显著差异(P>0.05);相关性分析结果显示,鸭SCD-1基因mRNA表达丰度与10周龄鸭血清中甘油三酯(TG)、总胆固醇(TCHO)、高密度脂蛋白(HDL)和低密度脂蛋白(LDL)呈显著正相关(P<0.05),与各常规肉质指标的相关性未达到显著水平(P>0.05),表明SCD-1基因在鸭的脂质代谢过程中发挥着重要的调控作用。

Abstract: In the present study,Cherry Valley ducks were subjected to diets with 4% linoleic acid and eicosapentaenoic acid (EPA) from week 2 to the end of week 10,respectively,and detected the mRNA expression of SCD-1 in 4,6,8 and 10 weeks,and analyzed its association with meat quality and serum biochemical indexes in 10 weeks.The Real-time fluorescence quantitative PCR results showed that the linoleic acid and EPA could significantly reduce the SCD-1 mRNA levels in liver,fat and breast muscles (P<0.05),meanwhile,the SCD-1 gene mRNA expression level had no significant difference between linoleic acid and EPA in every period (P>0.05).Correlation analysis showed that the mRNA expression levels of SCD-1 had significant positive correlation with serum TG,TCHO,HDL and LDL (P<0.05),and no significant correlation with all meat quality traits in 10 weeks (P>0.05).The research suggested that the transcription of SCD-1 gene was inhibited by linoleic acid and EPA,and played important role in the regulation of duck lipid metabolism.
