Sequence Determination of Full Genome and Analysis of the Gene Characteristics of Hankou/99 Strain of Foot-and-mouth Disease Virus

Abstract: Through sequencing and analyzing of full-length genome of Hankou/99 strain of foot-and-mouth disease virus (FMDV), the study identified type of the strain, which had enriched gene library of FMDV and provided a solid foundation for study of molecular variation, infectious molecular cloning and pathogenesis. Five gene fragments of S, L, C, D and E covering full-length genome of Hankou/99 isolate of FMDV extracted from infected cell fluid were obtained by RT-PCR technique, which were cloned and sequenced finally. The results indicated that full-length genome of the strain was 8099 bp including 5'NCR of 1040 bp ORF of 6966 bp and 3'NCR of 93 bp with 30 bp poly(A) at end of the genome. Through application of biological softwares, the strain could be ranked in O serotype FMDV. The strain had common characteristic of deletion of 30 bp in 3A region with the OLZ and TW/97 strains. In addition, the phylogenetic tree derived from the nucleotide sequence of VP1 gene from the strain and other nine reference strains showed that the strain presented higher homology with the four strains of OLZ, TW/97, O/Akesu/58, O/OMⅢ, which indicated that they belonged to the same genotype.
