Study on Serum-free Culture of Embryonic Germ Cells from Guanzhong Dairy Goat

Abstract: Goat embryonic germ cells (EGCs) was a kind of pluripotent stem cells derived from the primordial gonad, which isolation and culture system in vitro has an important value for the study of goat breeding. In this assay, Guanzhong dairy goat EGCs were isolated and cultured by enzyme digestion and tissue culture methods, and the effect of serum-free culture medium on cell proliferation in vitro was detected. The results were found that the medium could be used for obtaining of goat EGCs, and the colony morphology was typical, which expressed AKP, Oct4, TERT and SSEA-1. The EGCs could also differentiate into embryoid bodies, fibroblast-like cells, lipoblast-like cells and oocyte-like morphology by differentiation test in vitro. In conclusion, these data indicated that serum-free culture medium could be used in isolation and culture of goat EGCs, and our results provided a new reference to further establish the long-term culture system of goat EGCs.
