Study on Inhibition Effect of Broussonetial papyrifera Leaf Extract on Staphylococcus aureus and its Mechanism

Abstract: To investigate the inhibition effect of Broussonetia papyrifera leaf extract and its mechanism, the Staphylococcus aureus was used as test bacteria, the inhibition zone diameters were observed by the Oxford cup method, the growth curve, protein synthesis, and the membrane permeability of the the Staphylococcus aureus were detected to study the antibacterial mechanism of the Broussonetial papyrifera leaf extract. The results indicated that the antibacterial circle diameter of the Broussonetial papyrifera leaf extract and 75% ethanol extract were 17.5 and 18.0 mm respectively, the minimal inhibitory concentration (MIC) was 6.8 mg/mL, and the minimum bactericidal concentration (MBC) was 12.5 mg/mL. The results also revealed that the growth of the Staphylococcus aureus significantly decreased owing to the inhibition of the bacterial protein synthesis caused by the Broussonetial papyrifera leaf extract, but the membrane permeability was not affected. The Broussonetial papyrifera leaf extract showed obvious antibacterial activity on Staphylococcus aureus in vitro, and the antibacterial effect was probably associated with protein synthesis inhibition of the bacteria.
