Isolation and Culture of Amniotic Fluid-derived Stem Cells from Mongolian Sheep and Induction to Osteoblasts

Abstract: This study was designed to establish amniotic fluid-derived stem cells line and explore the potential to differentiate into osteoblasts. The amniotic fluid-derived stem cells were cultured under the condition of 15% FBS and detected the embryoid bodies forming ability and investigated the expressions of the stem cell markers (Oct-4,SH2,SSEA-1,Nanog,HLA-ABC,CD117) in different passage cell by RT-PCR. Furthermore,amniotic fluid-derived stem cells were induced by dexamethasone,β-glycerophosphate,ascorbic acid. Amniotic fluid-derived stem cells in vitro were able to expand extensively,form the embryoid body which expressed marker genes of three embryonic germ layers,and differentiate along osteoblasts. The results showed that we successfully isolated the Mongolian sheep amniotic fluid-derived stem cells and established cells line which can be induced to differentiate into osteoblasts.
