Establishment of a PCR-RFLP Method for Rapid Differential Diagnosis of Goose Parvovirus and Muscovy Duck Parvovirus

Abstract: A pair of specific primers was designed to amplify the goose parvovirus (GPV) and Muscovy duck parvovirus (MDPV) and establish a polymerase chain reaction and restriction fragment length polymorphism (PCR-RFLP) method for rapid detection of the infection status of waterfowl.An EcoRⅠ restriction enzyme was used which could digest MDPV PCR fragments into two different parts, with gene-lengthes of 530 and 280 bp, while GPV PCR fragments couldn't be digested by EcoRⅠ restriction enzyme. We established a fast differential diagnosis for GPV and MDPV using only one pair of primers and a restriction enzyme.
