Abstract: The antioxidative effect of melatonin (MLT) was evaluated on preserved bovine semen. Progressive motility (using visual observation), plasma membrane integrity (using the hypoosmotic swelling test, HOST) and acrosome integrity (using coomassie blue) of bovine spermatozoa were examined at 27 ℃ with 0, 10-3, 10-4, 10-5 mol/L concentration and for 1, 4, 8 h incubation time. The results showed that progressive motility, the percentage of plasma membrane integrity and acrosome integrity of the bovine spermatozoa were higher for 10-3 and 10-4 mol/L MLT than that of the control group (P<0.05). And progressive motility, the percentage of spermatozoa with plasma membrane integrity and acrosome integrity in melatonin (10-3 mol/L ) groups at 1, 4 h were higher than that in the control group (P<0.05). In conclusion, addition of optimal concentration of MLT helped to improve semen quality and life of preserved bovine semen.
