The Polymorphism of leptin Gene Intron 2 in Hu Sheep and its Influence on the Body Weight Traits

Abstract: Sequencing and PCR-RFLP were applied to detect single nucleotide polymorphism (SNP) in intron 2 of leptin gene in 85 Hu sheeps. Two SNPs of A99G and C150T were found. The dominant genotypes in the two loci were both wild genotype,genotype frequencies were 0.7882 and 0.8000, respectively. Mutation genotype was not found in the both loci. Chi-square test showed that the population was at Hardy-Weinberg equilibrium(P>0.05). Association analysis between the SNPs and body weight traits showed that the genotypes on the two loci had no significant influence on birth weight, 3-month weight and 6-month weight (P>0.05).
