单增李斯特菌ActA的表达、免疫原性研究及单克隆抗体的制备

Abstract

Abstract: The ActA of Lm was expressed in E.coli and the immunogenicity of ActA was evaluated. The monoclonal antibodies（McAbs） were prepared using the purified ActA as detection antigen. The subclasses, titers, affinity constants and specificities of the McAbs were detected. The GST-ActA was expressed in E.coli and the ActA inducted specific humoral immunity and cellular immunity. Four hybridoma cell lines against Lm were obtained, three of which were IgG1. The antibody titers of ascitic fluid of 3G6, 4E10, 2B9 were 1∶64000, 1∶64000, 1∶32000 and the affinity qualities were 6.62×10⁷ M^-1, 5.67×10⁷ M^-1, 7.15×10⁶ M^-1 respectively. The specificity analysis showed that 3G6, 2B4, 4E10 were specific McAbs against Lm and 2B9 was specific McAbs against Lm and L.ivanovii. The McAbs can be used to establish detection method of Lm.
