Codon Optimization of fat-1 Gene from Caenorhabditis elegan

Abstract: The objective of his study was to optimize the fat-1 condon from caenorhaditis elean. In this study, on the basis of no changing the amino acid sequence of fat-1 gene, we optimized the condon of fat-1 gene based on the codon preference of bovine muscle protein gene, in order to make fat-1 gene had high-level expression in cattle muscle. It could make a preparation for the production of fat-1 transgenic cattle. We changed 65 bases, which concering 65 codons and 7 amino acid. It accounted 5.38% for all the fat-1 gene bases. G+C contents increased from 45.08% to 50.04% and distributed uniformly, which was beneficial to the expression of the gene. The prediction result of the RNA secondary structure showed that the optimized gene could have high-level expression in cattle muscle.
