The Influence of Human Telomerase Reverse Transcriptase Gene for Goat Fetal Fibroblast

Abstract: Transferred eukaryotic expressing vector pCI-neo-hTERT into goat fetal fibroblast, screened and amplified the positive clones for RT-PCR detection, DNA ploidy analysis, cell cycle detection and apoptosis detection, investigated the influence of hTERT gene for goat fetal fibroblast. The 50th generation of positive clones expressed hTERT. DNA ploidy analysis showed the clones were normal diploid. The result of cell cycle of positive cells showed that compared with normal untreated cells (30 passages), the positive cells (50 passages) had higher S period, which illustrated these cells had strong ability to DNA synthesis and propagation. The result of apoptosis detection showed that the apoptosis rate of positive cells (50 passages) is extremely lower than normal untreated goat fetal fibroblast (30 passages). These experiences all showed that hTERT could increase the generation quantity of goat fetal fibroblast in vitro culture, maintain these cells a good morphology and strong propagation ability.
