Abstract: In order to study the basic effect of different serum on the construction of cell line, fibroblast cells were isolated from 20 seven-day old embryo of short-feet chicken and cultured, passaged and frozen via the method of adhere culture using 2 kind of FBS. The cell number, flask number, morphology and cell growth curve were observed. The results showed that various serum had significant effect on cell culture, and the cell characteristics were better when cultured with serum B than that with serum A. The present study provide the reference to the choice of serum on cell culture.
