Abstract: To found a method of getting much more high purity and high viability sertoli cells from adult rabbits’ testis，we used three enzymes which were collagenase,trypsin and hyaluronidase, digested with two steps, at last, isolated sertoli cells from adult rabbits. Then，we treated those cells with hypotonic shock in Tris-HCl, and through selected adhibition cultured, the sertoli cells were sublimated. At the same time,we used morphologic and HE dye to identify them. The result: we got plenty of sertoli cells which were high purity(78.3%)and high viability(90.7%).
