应用流式细胞仪研究阿非迪霉素对Jurkat细胞凋亡的影响

Study on Jurkat Cells Apoptosis Induced by Aphidicolin Using Flow Cytometry

摘要: 本研究应用流式细胞仪对添加了不同浓度阿非迪霉素的Jurkat细胞进行测定,通过Cellquest Pro软件分析细胞的凋亡率。结果表明,阿非迪霉素引起Jurkat细胞的凋亡呈剂量依赖效应和时间依赖效应。随着阿非迪霉素浓度的增加,早期凋亡率、晚期凋亡率和总凋亡率均不断增加,并在浓度≥1 μg/mL时,凋亡率呈明显增加的趋势;随着培养时间的延长,早期凋亡率、晚期凋亡率和总凋亡率同样不断增加,并在培养时间≥16 h,凋亡率呈明显增加的趋势。

Abstract: To study cell apoptosis changes induced by different concentrations of aphidicolin, we used flow cytometry to get Jurkat cell apoptosis rates. We found that dose and time dependent effects appeared because of Jurkat cell apoptosis induced by aphidicolin. With the increasing of aphidicolin concentrations or incubation time, the early apoptosis rate, late apoptosis rate and total apoptosis rate all increased. There was an obvious increasing trend when the concentration of aphidicolin was more than or equal to 1 μg/mL, or the incubation time was more than or equal to 16 hours.
