猞猁3类组织体外培养细胞的生物学特性分析

Biological Characteristic Analysis on Different Lynx Tissue Cells Cultured in vitro

摘要: 为研究体外培养猞猁细胞的生物学特性,深入开展猞猁基因组学及濒危野生动物保护工程,本试验获取猞猁不同组织细胞,用α-MEM培养液进行原代、传代培养,并对培养的细胞进行形态学观察、生长特性检测及核型分析。结果表明,猞猁不同组织细胞具有不同的生物学特性;心脏、躯体及睾丸来源的组织块分别在培养5~6、8~9、11~12 d时开始有成纤维样细胞从中长出,而在进行传代时,这3种组织来源的细胞生长速度没有表现出明显的差异,通常在4~5 d可铺满瓶底;核型分析结果表明,体外培养的猞猁细胞染色体数目为2n=38,为19对,其中18对为常染色体,1对为性染色体。上述结果可为今后猞猁不同组织细胞的深入研究提供试验材料和依据,也将为猞猁的细胞系构建提供参考。

Abstract: In order to study the biological characteristic on lynx cells cultured in vitro and do further development about lynx cell genomics with providing a reliable material platform for project of wildlife protection, different lynx tissue cells were got by tissue culture in this experiment, and α-MEM medium was used for the primary culture and serial subcultivation.Then morphology observation, growth curves and karyotype analysis were recruited for their biological characteristic analysis. The results showed that there were some differences between different tissue cells in lynx, ranging from morphosis to growth cycle. During primary culture the growth rate of different tissue cells showed differences.However, there were not significant differences among serial subcultivation.The further karyotype analysis indicated that the number of lynx cell chromosomes was 38, including 18 pairs of autosomes and one pair of sex chromosomes. These results provided experimental material and the basis for further research in the field.The study also would be a useful reference for the establishment of lynx cell line.
