猪繁殖与呼吸综合征病毒SYBR Green Ⅰ荧光定量PCR检测方法的建立

Establishment of SYBR Green Ⅰ Fluorescent Quantitative PCR Method for Detection of Porcine Reproductive and Respiratory Syndrome Virus

摘要: 根据GenBank中已登录的美洲型猪繁殖与呼吸综合征病毒(porcine reproductive and respiratory syndrome virus,PRRSV)在Nsp7基因中的保守区域设计合成了1对特异性引物,通过反应条件的优化,建立了检测美洲型PRRSV的SYBR Green Ⅰ实时定量PCR检测方法,同时与商品化荧光定量PCR试剂盒进行比对分析。结果表明,该方法特异性强,灵敏度高,能很好的区分美洲型猪繁殖与呼吸道综合征病毒和其他病毒。使用该方法对14份临床样品进行检测,阳性率85.7%,与商品化试剂盒符合率100%。

Abstract: A pair of specific primers conserved in the the Nsp7 genes of American porcine reproductive and respiratory syndrome virus (PRRSV) were synthesized. Optimization of reaction conditions, the detection of SYBR Green Ⅰ Real-time quantitative PCR method was compared and analyzed at the same time with the commercialization of fluorescent quantitative PCR kit. The results showed that the specificity of the method, high sensitivity, a good distinction between the American swine reproductive and respiratory syndrome virus and other viruses. 14 clinical samples were detected using this method, the positive rate was 85.7%, coincidence was 100% with commercial kits.
