Study of Pseudolaric Acid B Induced Apoptosis through ROS Regulating MAPK in MCF-7 Cells

摘要: 土槿皮乙酸B(PAB)通过活性氧诱导肿瘤细胞MCF-7凋亡,同时也通过调控有丝分裂原蛋白激酶(MAPK)家族蛋白来调控凋亡,但活性氧和MAPK的关系还不清楚,本研究探讨活性氧和MAPK家族蛋白的关系。4 μmol/L PAB在36 h诱导细胞死亡,活性氧清除剂抑制PAB的作用,并降低PAB的抑制率;同时,PAB促进JNK磷酸化,抑制ERK的磷酸化,此作用被活性氧清除剂抑制。

Abstract: The assay was aimed to study the relationship of ROS and MAPK family protein in pseudolaric acid B (PAB)-induced MCF-7 cell apoptosis. Contrast phase microscope was used to observe cellular morphologic alternation; MTT analysis detected the inhibitory ratio; flow cytometric analysis of DCFH-DA staining determined the level of ROS, and Western blotting detected the protein expression. We found that PAB induced MCF-7 cell apoptosis through increasing ROS and increased the phosphoration of JNK and decrease the phosphoration of ERK.
