高致病性猪繁殖与呼吸综合征病毒ORF5和Nsp2基因变异分析

Analysis in Heredity and Variation of ORF5 and Nsp2 Genome Sequence of PRRS Virus

摘要: 摘要 :采用RTPCR方法对2006-2007年华南地区部分省份的疑似猪繁殖与呼吸综合征(PRRS)阳性病料进行克隆和测序,获得13个Nsp2基因的部分序列片段和14个ORF5基因片段。序列分析结果表明,所获得的13个Nsp2基因序列中均有2个区域存在明显缺失,核苷酸的同源性为96.9%~99.1%,与中国代表毒株CH1a、美洲株代表毒株VR2332的核苷酸序列同源性分别为85.1%~86.4%和67.2%~68.9%,与疫苗株MLV的同源性在67.6%~68.9%之间,而与欧洲株代表株LV的同源性在54.1%~55.9%之间;获得的14个ORF5基因序列不存在缺失现象,核苷酸的相似性为同源性为98.8%~99.7%,推导编码氨基酸同源性为96.5%~99%。与国内毒株相比,核苷酸同源性为85.2%~100%,氨基酸同源性为84.6%~99%;与中国代表毒株CH1a、美洲型代表毒株VR2332的核苷酸序列同源性分别在94.4%~95.2%和86.7%~87.9%之间,与疫苗株MLV〖JP2〗的同源性在86.4%~87.6%之间,而与欧洲型代表株LV的同源性在54.1%~55.7%之间,推导编码的氨基酸序列同源性则分别在91.5%~93.5%,86.6%~88.6%,85.6%~87.6%,54.7%~56.2%之间。结果表明所获得序列的流行毒株均属于美洲型,进一步分析发现这些毒株可能均起源于同一个毒株,通过猪运输流通等途径在全国范围流行扩散。

Abstract: Abstract:Through RTPCR amplification of tissue or blood sample from some of pig form of Huanan area, 14 sequences of the ORF5 gene and 13 sequences of the partail Nsp2 gene were obtained. Sequencing and comparative analysis showed that two sequences exhibited deletion in the Nsp2 gene. There was 96.9% to 99.1% identity between those sequences of Nsp2 gene, sharing 85.1% to 86.4%, 67.2% to 68.9%, 54.1% to 55.9%, 67.6% to 68.9% identity with CH1a, VR2332 LV and MLV respectively. The identity among those sequences of ORF5 gene was 98.8% to 99.7%, sharing 94.4% to 95.2%, 86.7% to 87.9%, 54.1% to 55.7%, 86.4% to 87.6% identity with CH1a, VR2332, LV and MLV respectively. No deletion was exhibited in the 14 ORF5 gene sequences, and the sequences of ORF5 genes were analysed and compared with those of ATCC VR2332,CH1a, LV and MLV strains, there was 98.8% to 99.7% identity among those sequences of ORF5 gene, sharing 94.4% to 95.2%, 86.7% to 87.9%, 86.4% to 87.6% and 54.1% to 55.7% identity with CH1a, ATCC VR2332, MLV and LV respectively. And the homology comparision between the deduced amino acid sequence and other strains were 91.5% to 93.5%, 86.6% to 88.6%, 85.6% to 87.6%, 54.7% to 56.2% respectively. The results showed that the prevalent PRRSV strains in China belonged to North American in genotype, and the result showed that those virus probable origin of one strain, and prevail throught the transportation of the pigs.
