京海黄鸡禽流感抗病性状的全基因组关联分析

Genome-wide Association Studies for Jinghai Yellow Chicken's AIV Disease Resistance Trait

摘要: 试验旨在寻找影响京海黄鸡禽流感抗病性状的分子标记.本研究以京海黄鸡核心群母鸡为基础,测定了血清中的禽流感(H5亚型)抗体滴度,利用简化基因组测序技术进行全基因组关联分析(genome-wide association studies,GWAS),检测与禽流感抗体滴度相关的SNP位点.结果发现2个在染色体水平上与禽流感抗病性状显著相关的SNPs位点,分别位于4号和10号染色体上,其中10号染色体上的SNP位于RNA甲基转移酶家族基因Nsun7内部.该基因与细胞增殖和分化、蛋白质的生物合成密切相关,具有重要的生物学功能,推断其可作为影响京海黄鸡禽流感抗病性状的新的候选基因.

Abstract: In order to reveal the molecular marker that influence Jinghai Yellow chicken's AIV disease resistance trait.This research was based on female Jinghai Yellow chicken in core group and had measured the AIV antibody titer.Genome-wide association studies was carried out using specific-locus amplified fragment sequencing technology to detect SNP associated with this trait.Finally,two SNPs that reached the chromosome significant level with AIV were detected,they were on GGA4 and GGA10.The SNP on GGA10 was in Nsun7 gene and it had important biological function.Furthermore,we thought Nsun7 gene was an important candidate genes affecting AIV in Jinghai Yellow chicken.
