Cloning, Single Nucleotide Polymorphism and Bioinformatics Analysis of HSP70 Gene in Chicken

Abstract: This study aimed to clone the heat shock protein 70 (HSP70) gene, analyze its structure, and reveal the distribution of SNP in Wenchang chicken and Beijing-You chicken. HSP70 gene was amplified by PCR. Then the gene mutation was identified by sequencing. Three mutations were detected in the HSP70 genomic fragment, one in the coding region (A-1044G) and two in the 5′regulatory region (T-564C, A-68G). PCR-RFLP was used to analyze the polymorphisms of T-564C locus in our experimental chicken populations. The frequency of C allele was 0.87 in Wenchang chicken, 0.55 in Beijing-You chicken. Bioinformatics analysis of the promoter region had revealed that some transcription factor Sp1, AP-1, GATA-1, CREB, MZF1 and HSF2 were represented in this region.
