Extraction of Capsular Polysaccharide from Haemophilus parasuis SC096 Strain and Preparation of its Antiserum

Abstract: With the hot phenol method to extract the capsular polysaccharide (CPS), CPS was detected by SDS-PAGE and silver stain, and subsequently made emulsion with Freund's adjuvant.The antiserum was prepared by immunizing rabbits, and examined by agar diffusion test and indirect hemagglutination assay (IHA).The results showed that a high concentration of CPS was obtained by hot phenol method, SDS-PAGE electrophoresis showed that bands were diffuse distribution and the molecular mass was about 55 ku. Agar diffusion test result was positive and red blood cells occurred up to 10 hole drop degrees by IHA.The extraction of CPS and preparation of antiserum of Haemophilus parasuis SC096 strain provided a preparation method and the theoretical basis for a variety of other bacteria containing capsule, and made the foundation for study of antigenicity and vaccine of bacterial CPS.
