Molecular Cloning of Myf5 Gene and Biological Properties Analysis of Protein

Abstract: The aim of this paper was to amplify bovine Myf5 gene, and analyzed the protein biological characteristics. A pair of primers were designed according to bovine Myf5 gene sequences published in GenBank.Myf5 gene was amplified by RT-PCR from bovine back waist longest muscle, then linked to PUCm-T vector, restriction digested and sequenced. At the same time, we predicted the basic properties, the secondary structure and phosphorylated sites of Myf5 protein by the online tools. Having amplified coding areas sequence was 768 bp in length, encoded 255 amino acids. Protein biological characteristics analysis showed that Myf5 had the family genes typical basic helix-loop-helix domain. Serine(Ser) content was the highest in amino acid composition, and the phosphorylation site located in serine(Ser),threonine(Thr) and tyrosine(Tyr) residue.
