Research on Culture in Vitro and Identification of Bonamia ostreae

Abstract: Bonamiosis, caused by Bonamia ostreae, was a highly mortal disease of oysters and could result in enormous economic losses to shellfishery. Up till now, the study on the pathogen and the cause of the disease was much limited. To lay a foundation for the further pathogenic and diagnostic research of the disease, B.ostreae culture in vitro was preliminarily performed in this study. Based on PCR result, the B.ostreae positive oyster was selected and the tissue was separated for B.ostreae culture in vitro. Chosen in situ hybridization as a confirmation method, the results showed that the parasite could be detected after culture for one month. And the infection results determined by PCR assay and in situ hybridization were consistant. In this study,we established a simple and feasible culture method in vitro for B.ostreae.
