HA Gene Cloning and Sequence Analysis of Subtype H1N1 Swine Influenza Virus Isolated from Shandong

Abstract: HAgene of H1N1 subtype swine influenza virus was amplified by RT-PCR, the PCR products were obtained with the pMD18-T vector, and sequenced. Homology analysis showed that HA gene nucleotide homology of the isolated strain and other H1N1 subtype swine influenza virus were 70.7% to 90.8%, the highest homology was that with the A/swine/Zhejiang/1/2007, and homology of other strains were relatively low. Phylogenetic tree analysis showed that the evolutionary relationships of HA gene isolated in Shandong and that of European lineage of swine influenza virus were closet,proved that the isolated strain may be derived from the North American and Eurasian lineages of swine influenza virus lineage restructuring.
