Polymorphism Research of Somatostatin Gene in 13/17 Robertsonian Translocation Pigs

Abstract:

Karytype analyses were carried out in 309 pigs which were produced from three hybridized combinations of 13/17 Robertsonian translocation pigs by means of culturing whole peripheral blood.〖JP2〗 Three kinds of karyotypes appeared in their hybrids the normal karyotype pigs (2n=38), the heterozygous pigs (2n=37) and the homozygous pigs (2n=36). No mutation in exon 1 of porcine somatostatin gene was detected by means of PCRSSCP analysis in hybrids of three kinds of hybridized combinations while two single nucleotide polymorphisms (T520C,G521T) in 266~602 bp fragment of porcine somatostatin gene was detected by using the same method. But only two genotypes were detected: AA and AB. The result of genetic polymorphism analyses showed that the genotype frequency of AA(the limit of frequency was 0.58~0.77)and the allele frequency of A (the limit of frequency was 0.79~0.90)were both obviously higher than AB(0.23~0.42)and B(0.10~0.21) in hybrids of three kinds of hybridized combinations and pigs of different karyotypes. The genotype frequency of AB in the heterozygous pigs (0.42)were much higher than the genotype frequency of AB in the nomal pigs(0.25) and the homozygous pigs(0.40).The result of 2 test showed that genotype of all the populations reached HardyWeinberg equilibrium(P>0.05). 
