Study on Cloned and Purified FootandMouth Disease Virus by Plaque Technique

Abstract

Abstract: In this study, the AsiaⅠ_AKT03 strain of footandmouth disease virus(FMDV) was cloned and secondly cloned by plaque technique. The 50% of tissue culture infective doses (TCID₅₀) and 50% of lethal doses (LD₅₀) in suckling mice were compared with original, firstly cloned and secondly cloned FMDV.The results indicated that AsiaⅠ_AKT03 strain of FMDV could be successfully cloned by plaque technique. Through TCID₅₀ and LD₅₀ detection, it was found that the virulence of cloned strain was significantly higher than that of original virus. 
