Cloning of Bovine Leptin Mature Peptide and Bioinformatics Analysis before and after the Third Exon E3-125-C>A Point Mutation

Abstract:

In order to study the differences in the structure of the mature peptide of leptin gene before and after the third exon site directed mutation, and speculated its functional differences, the mature peptide of leptin based on GenBank was amplified using specific primers. And the mature peptide of leptin was cloned and sequenced. The physical and chemical properties, structure and function of encoding products of leptin were compared and analyzed by bioinformatics methods. Leptin single base mutations could change the encoding of amino acids, and result in a significant difference in the primary structure and secondary structure, and the tertiary structure was relatively stable. Single nucleotide mutations at the E3-125-C>A of leptin gene, caused structural changes that might cause functional changes.
