Detection of Porcine Epidemic Diarrhea Virus from 2011 to 2014 in Guangxi Provine and Sequence Analysis of their M Genes

Abstract: 331 piglets diarrhea fecal samples,collected from Nanning,Yulin and other 12 cites in Guangxi from January 2011 to April 2014,were used to detect porcine epidemic diarrhea virus (PEDV) by RT-PCR method.210 samples were PEDV positive,the positive rate was 63.44%.25 PEDV M genes were cloned from the PEDV positive samples.25 M genes were sequenced,analyzed and compared with 51 PEDV reference strains which were published in GenBank,and the phylogenetic tree was drew.The nucleotide and amino acid homologies of M gene were 96.0% to 99.9% and 94.3% to 99.6%,respectively.The genetic variation analysis of M gene showed that the PEDV prevalent strains of Guangxi province in 2013 to 2014 were genetically close to the prevalent strains of Beijing,Anhui,Wuhan,Hebei,Guangdong and other places of China from 2010 to 2013.But there were genetically different from the early Chinese isolates CH/S (GenBank No.JN547228),the vaccine strains CV777 (GenBank No.AF353511) and Attenuated DR13 (GenBank No.JQ023162).The results suggested that in recent years Guangxi PEDV epidemic strains had obvious variations compared with the early Chinese PEDV isolates.
