猪β2m在pMAL-p2X系统中的表达与纯化

Expression and Purification of Swine β2m Protein in pMAL-p2X System

摘要: 试验分别从pH、温度、菌体密度、IPTG浓度、诱导时间等条件对β2m在pMAL-p2X的表达进行优化,表达蛋白经过Amylose树脂柱进行纯化。结果发现,β2m在pMAL-p2X的最佳表达条件为:pH=7.5、T(温度)=37 ℃、D600 nm=0.3、IPTG=0.5 mmol/L和t(诱导时间)=5 h;在最佳表达条件下β2m的相对表达含量可达到45%;纯化后的融合蛋白纯度达到90%以上。

Abstract: Swine β2m was optimized to express in pMAL-p2X from several aspects including different pH value of the medium,induction temperature (T),cell density,concentration of IPTG and induction time (t),under the optimum expression condition,swine β2m was purified under amylose affinity resin column. The results showed that the optimum expression condition for β2m in pMAL-p2X was pH=7.5,T=37 ℃,D600 nm =0.3,IPTG=0.5 mmol/L,and t=5 h. Under the optimum expression condition, the relative expressed content of recombinant β2m protein reached to 45%, the purity of β2m protein was more than 90%.
