绵羊Cdkn1c的DNA甲基化分析

摘要: 为了探究绵羊Cdkn1c的甲基化状态,本研究对绵羊及其它3个物种的Cdkn1c mRNA序列进行了生物信息学分析,并通过亚硫酸氢盐测序法(BSP)对新生羔羊肾脏和肺脏Cdkn1c CpG岛上的CpG位点进行了甲基化分析。结果显示,整个编码区富含CG,为CpG岛;绵羊Cdkn1c所有分析位点均非甲基化,暗示该区域并非差异甲基化区域(DMR)。

Abstract: In order to determine the DNA methylation profile of sheep Cdkn1c, its mRNA sequences of sheep, cattle, human and mouse were subjected to bioinformatic analysis in the present study. Then bisulfite sequencing method (BSP) was used to examine the DNA methylation pattern of CpG island in Cdkn1c. The results of bioinformatic analysis exhibited that the coding region of Cdkn1c was a big CpG island in four species. And bisulfite sequencing results showed that all the analyzed CpG sites were completely non-methylated in the tissues of kidney and lung, suggesting that the analyzed region was not differential methylated region (DMR) in sheep.
