Screening of Membrane Proteins of Brucella abortus

Abstract: Application of immunoproteomic methods in research on Brucella abortus.Two-dimension electrophoresis was used to separate the membrane protein of Brucella abortus strain 544A grown under laboratory condition,and Western blotting was used to screen immunogenic proteins by serum of guinea and bov anti-Brucella abortus IgG.After ingel digestion,12 immunoreactive spots were identified by LC MS/MS.All the proteins were searched by the bioimformation tools,there were not only membrane proteins,but also cytoplasmic proteins.Most of them were involved in protein synthesis and metabolism.The immunoproteomic methods can be successfully applied in identifying immunogenic proteins of Brucella,which helps to search for other immunogenic proteins and lay a foundation for searching vaccine candidate antigens in future.
