Isolation and Identification of Reticuloendotheliosis Virus, Analysis of its Whole Genome Sequence and Pathogenicity Experiment

Abstract: Some pathologic samples were collected from chickens which were preliminarily diagnosed to be infected with reticuloendotheliosis virus (REV) in Guangdong province. The samples were inoculated on DF1 cells for reproduction. With identification of polymerase chain reaction (PCR) and indirect immunofluorescence assay (IFA), one REV strain was isolated, named as GD1210. According to the published sequences of REV, six pairs of primers were designed and synthesized. The overlapping fragments were amplified and the whole genome of GD1210 strain was sequenced. Sequence analysis showed that the full genome sequence of GD1210 strain was 8443 bp in size and shared the highest nucleic acid homology with the SNV strain when it was compared with the published REV sequences. The pathogenicity experiment for one-day-old SPF chickens of GD1210 strain indicated that the infected chickens had severe immunosuppression.
