Microdissection and LA-PCR Amplification of Swine Single 13/17 Robertsonian Translocation Chromosome

Abstract: The homozygotes of 13/17 Robertsonian translocation swine [2n=36,XY or XX,rob(13;17)] metaphase spreads were prepared from 3d culture of ConA-stimulated peripheral blood lymphocytes.The single 13/17 Robertsonian translocation chromosome was microdissected,and then amplified by the Linkeradaptor polymerase chain reaction (LA-PCR).After two rounds of PCR amplification ,smear DNA fragments ranging from 200~2800 bp were acquired. Microsatellite analyses (SWR1941 and SWR1120)and Southern Bloting confirmed that the LA-PCR production came from 13/17 Robertsonian translocation chromosome.The research will be useful for intensive molecular linkage map construction of chromosome 13 and 17, and the isolation of important genes located on chromosome 13 and 17.
