Effects of Acute Cold Stress on mRNA Expression of HSP60,HSP70 and HSP90 Genes in Altay Sheep

Abstract: In order to study the effects of acute cold stress on the cold resistance,production performance and disease resistance of Altay sheep by affecting the change of heat shock protein content.This experiment was set in the normal temperature group (15 ℃±2 ℃) and the cold stress group (-25 ℃±2 ℃),each group of 5,respectively.The heart,liver,spleen and kidney tissues were collected from the normal temperature group and the acute cold stress group after 24 h.The changes of HSP60,HSP70 and HSP90 mRNA contents were detected by Real-time quantitative PCR and homology analysis of HSP60, HSP70 and HSP90 genes.The results showed that the homology of HSP70 and HSP90 genes with the existing Ovis aries sequences were higher than 99%,and the HSP60 gene had more than 99% homology with the existing Bos taurus sequence.After cold stimulation,the expressions of HSP60 gene in heart,liver,spleen and kidney were higher than those in normal temperature group,and the expression of HSP60 gene in kidney was extremely significant different from that in control group (P<0.01).The expressions of HSP70 gene in heart,liver,spleen and kidney were higher than those in normal temperature group,and the expressions of HSP70 gene in liver and kidney were extremely significant different (P<0.01).The expressions of HSP90 gene in heart,liver,spleen and kidney were higher than those in normal temperature group,and the expression was significantly different in liver (P<0.05),but the expressions difference in spleen and kidney were extremely significant (P<0.01).It demonstrated that acute cold stress greatly stimulates the individual's heat production function,and changed its energy metabolism through the regulation of heat-producing related genes in the pathway,thus improving the survival rate of cells,enhancing the body's tolerance to environmental stress,and better adapting to the changes of environmental temperature.The results of this experiment provided a theoretical basis for further in-depth study of the effects of acute cold stress on Altay sheep.
