Effects of Matrine on intⅠ and dfrA of Integron in Clinical Multidrug Resistant Staphylococcus epidermidis Isolates

Abstract: To explore the effects of matrine on intⅠ and dfrA of integron in clinical multidrug resistant Staphylococcus epidermidis isolates.The K-B method was used to screen multidrug resistant isolates.The broth dilution method was applied to measure the minimum inhibitory concentration (MIC) and the best reversal condition.The ordinary PCR was applied to detect effect of matrine on intⅠ and dfrA gene sequences and Real-time PCR was applied to detect effect of matrine on intⅠ and dfrA mRNA expression levels.The ordinary PCR results verified that matrine had no effects on int Ⅰ and dfrA gene sequences.The Real-time PCR results demonstrated that int Ⅰ gene expression was down-regulated in presence of matrine rather than dfrA gene.Integron mRNA expression quantity of Staphylococcus epidermidis was decreased by matrine in vivo.
