Protective Effect of Sodium Selenite on Hepatocyte Apoptosis and the Decrease of Mitochondrion Transmembrane Potential Induced by Free Fatty Acids

摘要: 为研究亚硒酸钠对游离脂肪酸 (free fatty acid,FFA) 致体外培养大鼠肝细胞凋亡和线粒体膜电位降低的保护作用,本试验通过胶原酶两步灌流法分离培养肝细胞,在细胞培养液中添加FFA和亚硒酸钠 (0.1 μmol/L)测定细胞的凋亡情况、细胞功能和线粒体膜电位。结果表明,随着培养液中FFA浓度的升高,细胞凋亡加重,肝细胞功能受到影响,线粒体膜电位降低且FFA的这种作用呈剂量依赖性;亚硒酸钠能抑制由FFA引起的肝细胞的上述变化。

Abstract: The objective of this study was to define the effect of sodium selenite on hepatocyte apoptosis and the decrease of mitochondrion transmembrane potential induced by free fatty acids (FFA). Primary rat hepatocytes were obtained with two-step collagenase perfusion method. We estimated hepatocyte apoptosis, liver function and mitochondrion transmembrane potential in FFA treated group with or without sodium selenite (0.1 μmol/L). Our data indicated that FFA changed dose-dependently hepatocyte apoptosis, liver function and mitochondrion transmembrane potential, whereas the presence of sodium selenite reversed these alternations.
