猪繁殖与呼吸综合征病毒的分子流行病学分析

Analysis of Molecular Epidemiology of Porcine Reproductive and Respiratory Syndrome Virus

摘要: 为研究中国猪繁殖与呼吸综合征病毒(PRRSV)的基因变异情况,对GenBank中公布的中国不同时间和地区分离的53株PRRSV毒株的N、GP5基因序列分别进行了遗传变异分析。根据N基因构建的遗传进化树分析表明,所有的中国大陆分离株均属于美洲型(NA),中国的PRRSV分离株可分为4个亚群,亚群1中的毒株主要分布在中国东南部地区,其GP5主要中和抗原表位基因高度变异;亚群2主要为以CH1a株为代表的毒株;亚群3中的毒株与疫苗株MLV和VR2332高度一致;而台湾地区的分离株与大陆地区的分离株差异较大,单独归属于亚群4。这些研究结果对了解中国流行的PRRSV毒株的分子流行病学具有重要意义,也可部分解释MLV或灭活的CH1a疫苗对亚群1病毒感染猪保护力较低的原因,为今后制定PRRS的防控措施提供科学依据。

Abstract: To study the genetic variation of PRRSV isolates in China, we compared the N and GP5 gene sequences of 53 strains of PRRSV isolates which were isolated from 1996 to 2008 in China, and had been published in GenBank. The cladogram according to the N gene demonstrated that most of the Chinese isolates were belonged to North American type. Among them four highly diverse subgroups were clearly demarcated on the NA-genotype phylogenetic tree. All of the subgroup 1 isolates restricted geographically to regions of the southeast of China were found to be highly variable in the primary neutralizing epitopes. The first PRRSV isolate (CH1a) was of the representation of subgroup 2. Subgroup 3 isolates were shared high identity with MLV vaccine and its parent virus VR2332. The strains in Taiwan were belonged to subgroup 4, which were different from those in mailand China. These results may contribute to the knowledge of PRRSV epidemiology in China, and may help to explain the low efficiency of MLV or inactivated CH-1a vaccine to protect pigs infected the subgroup 1 virus, and the great genetic diversity should be taken into consideration for control and prevention measures. 
